If you are talking about the quick disconnect style of fittings (no threaded connections) the fitting is removed by pushing on a circular ring around the tube at the fitting connection. You can buy a special plastic ring release tool from Harbor Freight or most auto part stores fairly cheap. The ring tool snaps around the metal transmission line, then slides up to connector fitting. By pushing on the fitting release ring, you are releasing the tension of the spring fingers on the ring, allowing the fitting to be pulled apart. It can be tricky, you may have to push the fitting together to allow the spring fingers to "un-bite" the connection while you are pushing the release ring at the same time. Once it releases, it comes apart so easy, you wonder why it looks so difficult to remove. Go check out a Plumbing section at Home Depot and look at the "Sharkbite" style quick fittings--they are the same design and disconnect using the same principal.
if you are asking about the cooling lines for the transmission that connect to the radiator, its easy. first disconnect the lines from the radiator (2 lines, usually on the bottom opposite the radiator outlet) then disconnect the lines from the transmission. remove the brackets holding them on, then remove.
there are two metal looking lines that screw into the radiator that is the cooling side
Remove fan shroud Drain radiator Disconnect upper and lower hoses Disconnect transmission lines if Auto Remove securing bolt at top and side Radiator should lift from lower supports
Drain the old one, disconnect the transmission lines, undo the radiator hoses and bolts holding it in an pull it straight up.
remove the metal plate that covers the radiator, if you ever change a headlight blub, you already know which plate im speaking of... afterwards after disconnecting hoses transmission a/c lines just pull upwards, oh and disconnect plug to cooling fans it all comes out together ( fans, radiator, transmission cooler)
There is a special tool that releases the wire clips that hold them on.

If there are lines coming from the transmission to the radiator, it has a cooler in the radiator. If there are lines from the transmission to an external heat exchanger, that would be an auxiliary cooler.
there is a tool made to slip around the line rite at the connectionto the radiator and you push the tool in and pull the line. i believe its called a compression line remover or a fuel line remover. most auto parts stores have them.
The transmission cooler lines are located coming from the radiator on the bottom. One is for the sending and the other for return. The lines are high pressure, just as your radiator hoses are when the car is running. The lines go from the radiator to the top of the transmission.
Vehicles with an automatic transmission may have lines that carry transmission fluid from the transmission to a separate tank on the bottom of the radiator.
