To check the timing, the tan/black stripe wire to the distributor needs to be unplugged. Use # 5 plug wire with the lower timing mark. The factory timing should be labeled on the air cleaner. It is probably 4 degBTDC but 6 to 8 degBTDC should make it run cooler and give it better gas mileage.
Check if you have secondary timing marks installed. Mine are bolted to the oil pan and easily accessible from the bottom. Instead of using cyl #1 for setting the timing, you will have to use #5 or #8 (to make up for the 90 degrees offset to the factory timing marks

the timing marks are on the rear of the engine @ the top of the transmission bell housing,use the mark on the flywheel & pointer on the bell housing to line up the timing marks.

The 2000 Ford 4.2 engine does not have any timing marks on the crankshaft. Instead, you have to use a timing kit to install a new belt.
if you mean the ingintion timing marks, they are on the crankshaft pully. Use a rag and some cleaner to see them clearly.
I believe what you're talking about is that there is two timing tabs on the motor. One at 1 o'clock and 1 at 5 o'clock. The top one is hard to see so gm put one on the bottom where it is easier to get to or see. If you use the top tab you hook your timing light to the #1 plug wire and if you use the bottom tab you hook to the #5 wire
The one on the top is for the first cylinder, the one on the oil pan is for easier access, use cylinder #5 or #8 on it.
When you say 'Timing Marks', there are several places on this engine that has various marks in regards to the Harmonic Balance, Camshaft sprockets, crankshaft, and timing cover. If you are checking the timing, you will use the marks on the timing cover. If you are setting or replacing the timing belt, you will want to use the camshaft sprockets in relation to the crankshaft/harmonic Balance. On this year, the crankshaft key notch should be 12'oclock and the sprockets should have the timing marks 'E' for exhaust and 'I' for intake aligned facing with the 'E' at 3'oclock and the 'I' at 9'oclock. no you need timing tools put a timing pin in the block to lock the crankshaft and use the timing tool to lock the cams
