Iran, Iraq and Jordan are passable, but your passport is going to have to be acceptable to all of them. I don't think as USA passport will work that well.
The African leg is well trodden. Overland tourist trucks do it every weeks. The roads are all car-passable. The tour companies just use trucks to look butch - a normal luxury bus would be more comfortable.
Egypt has good roads - and huuuuuge piles of red tape for vehicles in transit through Egypt. The problem is Sudan - due to the war in southern Sudan you have to travel on the Nile ferry. So no road transport allowed.
From Kenya south through Tanzania, Malawi. Zambia, Botswana and South Africa there are reasonable to very good roads.
