Extension(70oC): this is the portion that replicates the DNA

These step repeat, each time constitutes a cycle. The number of DNA replicated = 2n where n equals the number of cycles

The basic steps of the PCR reaction are:

  • Denaturation - the parent DNA molecule is separated so the individual DNA strands are exposed
  • Primer annealing - the oligo nucleotide primers bind to the parent DNA at specific locations
  • Primer extension - A thermo stale DNA polymerase extends these primers and forms the daughter strands
  • Cyclization - The process is designed to repeat in a cyclical manner for 30 or more cycles
