The quickest route is through Niagara Falls and then on down towards Erie, PA on the interstate.
Define "better." They're different sections of I-75, so it's mostly interstate driving. Fort Lauderdale to Naples is better known as "Alligator Alley." It's shorter and faster… than the Tampa route, but other than a few tourist traps, there's a whole lot of nothing to see. Tampa to Naples is a coastal route and you're usually near the water. The Corkscrew Swamp Sanctuary is a must-see for nature lovers.
