Thin layer chromatography can be used to identify a compound because for certain reaction mixtures, TLC can separate all compounds in a mixture based on the distance the compound travels on the TLC film. With the proper solvent system, TLC can be used to uniquely identify a compound.

Chromatography uses a phenomenon called partition coefficient to separate mixtures which are dissolved in a liquid or gas and then passed through a stationary phase. This stationary phase slows down different constituents of the mixture by different amounts, so they are separated.
Identification is undertaken either by combining a gas chromatograph with a mass spectrometer, which can identify compounds as they exit the chromatography column, or by producing a chromatograph of a known mixture with that of an unknown mixture. Compounds which have the same "retention time", under the same conditions, are likely to be the same.
