Highway 16 is central and northern Alberta's primary east-west highway.

It is part of the Yellowhead branch of the Trans-Canada Highway system that extends from British Columbia to Manitoba. Within Alberta, it passes through Lloydminster, Vermilion, Vegreville, Elk Island National Park, Edmonton, Edson, Hinton and Jasper National Park.

The length within the province of Alberta is 641 km (398 mi).

It is a free-flowing four-lane divided highway for the majority of its stretch through Alberta except for the section from west of Hinton through Jasper National Park to the BC border—future plans for further westward twinning through this section are unknown. The highway currently passes through signalled intersections in Hinton, Edson and Lloydminster, and bypasses around these major centres have been discussed.

Within the city of Edmonton, the highway forms one of the main east-west routes and is known as Yellowhead Trail. Most sections of Yellowhead Trail have gradually been upgraded to a free-flowing freeway, but signalled intersections still remain in some heavily commercialized sections. Future plans call for replacement of all signalled intersections with interchanges, flyovers and closures.
