U.S. Route 25E is the eastern branch of U.S. Route 25 from Newport, Tennessee, where US 25 splits into U.S. 25E, and U.S. Route 25W, to North Corbin, Kentucky, where US 25E rejoins US 25W. The road, however, continues as US 25E for roughly 2 miles (3.2 km) until it joins Interstate 75 in North Corbin. The route, included in the original 1926 map of the U.S. Highway System, passes through Morristown, Tazewell, Harrogate, Middlesboro, Pineville and Barbourville.

The highway passes through the Cumberland Gap Tunnel at the Tennessee-Kentucky state line.

From Interstate 81 north of White Pine, Tennessee, to Corridor F (State Route 63) in Harrogate, US 25E is Corridor S of the Appalachian Development Highway System.^[3]

In 2009 the Tennessee stretch of 25E, extending from Newport to Cumberland Gap through Cocke, Jefferson, Hamblen, Grainger, and Claiborne Counties, was designated a National Scenic Byway called the East Tennessee Crossings Byway.^[4]
