What is

dornase alfa inhalation solution?

DORNASE ALPHA helps to manage lung problems caused by cystic fibrosis. It helps loosen the mucus in the lungs so that they can function better. It also decreases the number of lung infections that require treatment with intravenous antibiotics.

What should I tell my health care provider before I take this medicine?

They need to know if you have any of these conditions:
• an unusual or allergic reaction to animal proteins
• an unusual or allergic reaction to dornase alfa, other medicines, foods, dyes, or preservatives
• pregnant or trying to get pregnant
• breast-feeding

How should I use this medicine?

Do not use this medicine until you have been trained in the proper technique. This medicine is for inhalation only using the nebulizers and compressors recommended by the manufacturer. Contact your doctor or health care professional if you are not sure if you have the correct nebulizer and compressor. Read and follow the instructions provided to you carefully. Do not use a mask. Use the mouthpiece provided with each nebulizer kit. Wash hands before and after use. Consult your doctor or health care professional if you are having problems with or have questions about the proper inhalation technique or the use, maintenance, or storage of your nebulizer and compressor.

Contact your pediatrician or health care professional regarding the use of this medicine in children. Special care may be needed.

What if I miss a dose?

If you miss a dose, use it as soon as you can. If it is almost time for your next dose, use only that dose. Do not use double or extra doses.

What drug(s) may interact with dornase alfa?

There have been no interactions with other medicines reported to date. Do not mix any other medicine in the nebulizer with this medicine.

Tell your prescriber or health care professional about all other medicines you are taking, including non-prescription medicines, nutritional supplements, or herbal products. Also tell your prescriber or health care professional if you are a frequent user of drinks with caffeine or alcohol, if you smoke, or if you use illegal drugs. These may affect the way your medicine works. Check with your health care professional before stopping or starting any of your medicines.

What should I watch for while taking dornase alfa?

Visit your doctor or health care professional for regular checks on your progress. Follow your doctor's instructions carefully.

Keep all other inhaled medication systems completely separate from this medicine.

What side effects may I notice from using dornase alfa?

Side effects tha you should report to your doctor or health care professional as soon as possible:
• allergic reactions

Side effects that are generally mild, do not require medical attention and go away within a few weeks:
• chest pain
• eye irritation or infection
• hoarseness
• pharyngitis, laryngitis
• rash

Where can I keep my medicine?

Keep out of the reach of children.

Store in a refrigerator between 2 and 8 degrees C (36 and 46 degrees F). Protect from light. Do not expose to room temperature for more than 24 hours. Do not use if solution is discolored or particulate matter is present. Once opened, the entire ampule must be used or thrown away.

Dornase alfa (proprietary name Pulmozyme from Genentech) is a highly purified solution of recombinant human deoxyribonuclease I (rhDNase), an enzyme which selectively cleaves DNA. Dornase alfa hydrolyzes the DNA present in sputum/mucus of cystic fibrosis patients and reduces viscosity in the lungs, promoting improved clearance of secretions. This protein therapeutic agent is produced in Chinese hamster ovary cells.

Dornase alfa is the most recent therapeutic agent developed with this basic mechanism of action. Prior to the cloning of the human enzyme, bovine DNase I was on the market for many years, though its utility was limited by the inherent antigenic response to a cow protein in the lungs of patients. Other DNases, such as DNase II, have therapeutic potential as well, but as of yet no further DNases have been brought to market for cystic fibrosis.
