There are over 150 interchanges, numbered from west to east, on Highway 401 in Ontario. Originally, the freeway had used sequential exit numbering, with roughly 128 numbered exits, before switching to the milemarker system, which in turn led to the present kilometer-based system after Canada's highways adopted the metric system.

Proposed extension into Michigan
As part of the Detroit River International Crossing project, the following interchanges are planned on a western extension of Highway 401 to the Ambassador Bridge. They are only conceptual, and vary considerably between alternatives; the below is a rough combination of all alternatives[5]. This concept plan was put forth by the Detroit River International Crossing, with significant input from the Windsor City Council, and has been named the "GreenLink" proposal.[6]
However, on May 1, 2008, there has been a proposed extension of Highway 401 which would be named the Windsor-Essex Parkway. It would run parallel alongside the Highway 3 going north and going west along the E.C. Row Expressway towards a new inspection plaza and river crossing in West Windsor. This project will cost about $1.6 billion but the parkway will put heavy trucks off local streets and there will be several tunnels so local residents can cross over the parkway with ease.[7]

Other planned interchanges
The reconstruction of Highway 401 in Mississauga is expected to add several interchanges in order to serve that fast-growing region.
Another widening project throughout Oshawa will see old interchanges closed in favour of newer ones. Construction was completed on a new interchange (exit 415) at Stevenson Road in Oshawa. This upgrade with new ramps and bridges will provide easier access to the General Motors (Canada) plant and the Oshawa Centre Mall. It replaced the outdated Park Road interchange (exit 416) that was be closed off in late May 2009.
After 407 ETR
gets extended east to Highways 35/115, two other freeways will be built linking the 407 and the 401 together;
- 407-401 Durham West Connector, near the boundary of Ajax and Whitby,
- 407-401 Durham East Connector, in Clarington.
An interchange is in the planning stages for Lakeridge Road (Durham Road 23) once Highway 401 continues to widen east of Ajax with an Environmental Assessment and when the Durham West interchange is in place.
A new interchange at Wonderland Road in London is planned to be built around 2015.
