  1. The technique of measuring the light absorbed, reflected, or emitted by a microscopic specimen at different wavelengths.
  2. Such a technique used to characterize and quantify nucleoproteins in single cells or cell organelles on the basis of their natural ultraviolet absorption spectra or following stoichiometric binding in selective cytochemical staining reactions.
