a method for investigating protein-protein interactions in which a protein of interest is expressed as a fusion protein with glutathione-S-transferase (GST) or an epitope tag such as 6 × His or c-Myc. This is used to explore previously unknown interactions with other proteins in a mixture such as a cell extract. Protein complexes formed with the fusion protein are precipitated either with specific antibodies or with a suitable affinity adsorbent, and the nature of the bound proteins is identified by gel electrophoresis and mass spectrometry.
