U.S. Route 601 is a spur of U.S. Route 1. It runs 318 miles (512 km) from Mount Airy, North Carolina at U.S. Route 52 to U.S. Route 321, near Tarboro, South Carolina.

In North Carolina, U.S. 601 is concurrent with Interstate 85 between Salisbury and Concord, a distance of approximately 17 miles (27 km). In South Carolina, U.S. 601 shares 5 miles (8.0 km) of highway with its parent route, U.S. 1, near Camden.

South Carolina

US 601 begins at US 321 near Tarboro, SC and intersects with such highways as US 278 in Hampton, US 301 between Bamberg and Orangeburg, US 78 in Bamberg, I-26 near Orangeburg I-20 near Lugoff, US 1 in Camden, US 521 between Camden and Kershaw, and SC 9 in Pageland.

North Carolina

After crossing the North-South Carolina border, US 601 intersects with such highways as US 74 in Monroe, NC 24-27 in Midland, NC 49 south of Concord, I-85 near Salisbury, I-40 near Mocksville, US 421 in Yadkinville, I-74 near White Plains, and US 52 in Mount Airy.
