The university was founded by an 1860 decree of Domnitor Alexander John Cuza, under whom the former Academia Mihăileană was converted to a university.

Corneliu Zelea Codreanu had been a student at Iaşi until his expulsion in 1922. In June 1923, poor university conditions and the modification of Article 7 of the new Romanian Constitution to allow immigrants, including Jews, to gain citizenship, led to student strikes throughout Romania. The National Christian Defense League, founded by Codreanu and Professor A. C. Cuza, took over the university and flew its swastika banner over the entrance. These events were leter investigated by a committee headed by Rector Traian Bratu, Constantin Rădulescu-Motru, and Nicolae Basilescu.
