The transmission must be removed to access the two freeze plugs on that side. It is easier to remove the engine, but techinically, no, you do not _HAVE_ to.
I have never replaced one this model but in order to change a freeze plug you really need to pull the entire engine out and usually many of the accessories on the engine in order to pull it out with a channel lock or similar tool. than simply tap a new freeze plug in.

I had an 89 Topaz and I had to replace 3 freeze plugs. It had a four cylinder and 2 were on the back side (closer to the firewall). The hard part was getting it out. I used various tools including screwdrivers, pipe with hammer to just get the plug out. Because of limited space I went down to Autozone and they had a freeze plug with a piece of rubber between to metal washers with a bolt going through the middle. You simply put it in the hole and tightened it with a rachet. I started realizing that I had been putting more water than antifreeze in the radiator causing them to rust out. The last one to rust out when between the engine and transmission. In order to get to that you had to take out the engine!! No thanks not for a 25 cent piece of metal. The car had 180K so I just sold it for scrap and bought a 94. A hard lesson learned.
