Cats spray to communicate and they communicate in different ways so a cat will spray for different reasons.
The main reason that they spray is because they are not spayed or neutered. Spaying or neutering your cat helps in mostcases of spraying (both male and females can spray).
If your cat is fixed and STILL sprays it is either marking its territory or stressed.
Cat spray contains pheromones, which sends messages during mating seasons. They also spray to mark territory, letting other cats know that this is their area. If you have a lot of cats they will spray more.
The best way to get them to not spray a certain area again is to throughly clean the area. There are certain cleaners that can be purchased at pet supply stores that disinfect and deodorizer a cat spray and urine smell. If a cat smells a spray spot he may spray there again. A equal mix of white vinegar and water in a spray bottle will work too.
Another option they sell is "Feliway" which helps calm a cat.
Pet stores like PetSmart also sell something called "SSScat" if they are spraying in a certain area SSScat is a motion detector that will spray a very harmless mist when the cat gets near where it is placed. This will keep them from returning to the area.

Is he trying to get into the room or does he just like to scratch? If he's trying to get in, let him in. If you don't want him in the room, ignore the scratching and he will eventually see that it's not working and stop. This works short term and long term, but you need to be consistent. If he just likes to scratch, spray him with some water and he will learn to stop
Spray it in the face with water or spray deterrent on the rugs. If you haven't litter trained it yet, that's obviously the problem.
