Once you've downloaded it, all you need to do is start the game at the start menu as you normally would. When you load the game, it will jump to a short cutscene of Rebecca talking about how she had fixed some of Ezio's memories. Once the Animus loads, there should be two new mission symbols on your mini-map. Go to them to initiate their content.
Note: You shouldn't play the Da Vinci Disappearance until you completely finish the Brotherhood campaign, as it contains spoilers.
