It depends on which vehicle you are speaking of. There are usually specific instructions in your owners manual for this. It usually involves using the ignition switch and your remote.

Check the fuse and relay.
There are four ways to program most of these locks; refer to your owners manual for the proper sequence.
disconnect the fuse that works the locks.
The power door locks on a Dodge Caravan can stop working if the fuse is bad. Check the power door lock relay fuse that is located in the engine compartment on most Dodge Caravans. It should be next to the battery.
Take key from ignition and lock the drivers door, then unlock it. Another way is to put the key into the trunk lock and turn it. Stop locking the car by depressing lock when you exit. Use the key or automatic door locks.
The problem is that your door locks / latches are sticking. Spray some WD-40 on all the door jams/locks including the rear door hatch. The problem will stop.
doors locks & trunk lock not working
I had a situation with my elect. door locks going and going no stop while driving. I went to Jeep dealership and found out that a module is in the passenger door panel which operates the locks and windows & security system. Also another one on the drivers side. They disconnected the one on the passenger side and the problem fixed. but can't operated windows and lock doors manually. My drivers door still locks auto. the new module was $389.00 from Jeep plau labor so ...were talking about $800.00 plus for this repair from dealership.....I don't think so.
I had all four of my door locks stop working. I was told it was the actuators and for $250.00 per actuator (my only local Ford dealership) I could have them fixed. Back to manual for me.
you have a short some where on the door wiring take the door panel out and get a DVOM and test it or else bring it to a local shop and get it test it
