To unlock new characters, you have to complete missions with specific characters and you can also play alot of versus matches as well as buying them from the shop with the ryo you earn. Ryo is aka money;

WARNING, ANSWER IS VERY LONG.First, make sure you beat story mode on Attack power 2 Difficulty 2 (No more, no less.) after that is done, buy all the characters you can, ionce you have bought all of the characters from the first set(s), Kabuto, orochimaru, and sasuke will appear in the shop.(Including Sai and Yamato, I think.) A long story short, just keep buying all the characters and modes and stages. At the end, after you get kakuzu and hidan, you will have to do something else for bando and anbu kakashi. so, just keep buying! SORRY THIS ANSWER WAS SO LONG.(Sasuke is 45,000 ryo.)

EDIT TO ANSWER: Yeah, but the prob is, Anbu Kakashi costs 100,000 ryo lol... and deidara is like impossible to beat on Difficulty 2 Power 2 (Took me FOREVER) also once you do buy all characters i think something special happens... Also when you DO try and unlock Anbu Kakashi, make sure your at 40% mission completion, plus, that you beat his S-Rank mission on any rank higher than Special Jonin, after 40%, go back and do the S-Rank again, i know this got nothing to do with sasuke but hey, every bit helps to get characters
