∙ 14y agoDifferent microbes grow best at different temperatures. If you are looking for a particular microbe, you should incubate at the best temperature for that microbe, which may not be the best for a different microbe. If you just want to see what will grow, you will need to incubate at several different temperatures.

∙ 11y agoThe temperature at which agar plates are incubated has nothing to do with agar. The temperature of 37 degrees celsius is used because it is the optimum temperature for most bacteria like E. coli, Staphylococcus aureus, etc. Agar plates are incubated at lower temperatures also. One such example is the growth of yeast (Saccharomyces cerevisiae) and the plates are incubated at around 25 degree celsius. It all depends upon the optimum growth temperature of the organism streaked on the plate.
