Does mass measurement include the effect of earth's gravitational pull on the object being measured?

Mass is not affected by gravity. When you measure the mass of an object with a balance, you would get the same result under any gravitational conditions except for under micro- or zero gravity, in which case there would be insufficient force to move the balance to equilibrium.
