No, there is no connection between losing transmission fluid and the speed odometer not working. My mother's car lost all of it's transmission fluid but the odometer still worked.

If both odometer and speedometer are not working I would blame the vehicle speed sensor. If just the odometer isn't working the gauge cluster assembly needs to be replaced with a reconditioned one.
