No. The first European settlement in what is now the United States was in what is now Pensacola, Florida (settled in 1559). The first successful permanent settlement was in what is now St. Augustine (settled in 1565). North Carolina was not settled until the 1580s (and both of those settlements failed).

General consensus is that Virginia is indeed the first "colony" with the establishment of Jamestown in 1607.

This was followed by Massachusetts (Plymouth and Massachusetts Bay Colony) in 1620 and New York (originally New Amsterdam) in 1626.

I am assuming that you meant American colony, as Jamestown was certainly not the first colony in world history.
