The three main factors that affect enjyme activity are:- a)Temperature:Enzymes generally function properly at a narrow rangr of temperature and shows its highest activity t a particular temperature called its optimum temperature. b)pH:-as temperature pH also has a particular value ,in which the enzymes are most active,called the optimum temperature. c)Concentration of substrate;-with the increase in substrate concentration,the velocity of the enzymatic reaction also increases,till a maximum velocitywhich does not rise any further with the rise in substrate concentration.

Third factor is the pH value.Enzymes can work efficiently over a narrow range of pH called as Optimum pH.A minor change in pH value can denature the enzyme.

Whereas the 5'-3' exonuclease activity is an also repair strategy exercised by the DNA polymerase I. However, in this case the polymerase would move in the forwards direction and excise the miss-matched nucleotides at any position regardless with one nucleotide far or so many. After a certain level of enzyme concentration, there will be no more increase in the enzymatic activity because all the substrate molecules are combined with an enzyme and the rate of reaction will stabilize.2: Substrate concentrationBy increasing substrate concentration, enzymatic activity increases. Increasing the substrate further without increasing the enzyme concentration will not affect the enzymatic activity because all the enzymes are occupied by a substrate molecule.3: pH valueSome enzymes require acidic surroundings, most require a more neutral condition for their activity. Change in the pH can change the enzyme's structure and enzyme become useless.4: TemperatureAn increase in temperature of 10 degree celsius doubles the enzymatic activity. If the enzyme denatures, the substrate can not fit in to the active sites and enzyme become useless.
