The largest building in New York City is the MetLife Building in Midtown, at more than 3.1 million square feet. At the time it was built, it was the largest commercial property in the world. Note that it is not the tallest building. That distinction belongs to the Empire State Building, at least until the new One World Trade Center (a.k.a. Freedom Tower) is completed.

The second largest building is the Port Authority building in Chelsea at a little under 3 million square feet. Google recently purchased the building, in which it had been leasing space for its New York Headquarters.

One of the very oldest building in NYC is a small house that was used by one of Dutch settlers in the 1600's. I am not sure where it is located in NYC, but have seen pictures of it and know that it is open for visits.

Fraunces tavern is the oldest building in New York city, located near the southern tip of Manhattan built in 1719. St. Paul's chapel is the second oldest building constructed in 1764 and is located near wall street in lower Manhattan. St. Paul's is so old that George Washington worshipped there. Finally, the Morris-Jumel Mansion is located in northern Manhattan in an area called Washington heights and was constructed in 1765.
