The 1st Amendment guaranteed the freedom of religion. In fact, in guarantees both the freedom to establish religion and the freedom to practice religion. Other rights guaranteed in the 1st Amendment include freedom of speech and freedom of the press.
The first amendment of the constitution does guarantee the freedom of religion.
They are separate amendments. Freedom of speech and press is the 1st amendment. The second amendment is the Freedom of religion.
There are two clauses dealing with freedom of religion in the Constitution of US. First one is Establishment Clause and the second one is Free Exercise Clause.
The first amendment.

The First Amendment of the U.S. Constitution protects the right to freedom of religion and freedom of expression from government interference.
The First Amendment prohibits the government from establishing a religion, which by implication prevents its either supporting or inhibiting religion in general.
