The person you're trying to send a message to has to be following you.
On Twitter, you can either send them a message while you're publicly updating (eg. "@Skunk_munchies your plugging yourself!!") or you can click on "direct messages" which is in the menu to the righthand side of the page. You can only send a direct message to people who are following you by either clicking: * the "message" link on the profile page * the reply icon attached to messages in your direct message inbox * the drop down box on the direct message inbox page * the status update box using the direct message command: d + username + message (eg. "d skunk_munchies stop plugging yourself while answering a question")
On Twitter, in the top right corner, there is a small picture of a person and an arrow, click on that, and then in bold, there is an option that says Direct Messages. Just compose one to somebody. Either that or you could go on somebody's profile, and then I think there is a 'Message' option.

When logged in to your account, the Twitter homepage will have a section that says: "Direct Messages". Click on this link and from here you are free to message your followers. Or, you can find someones user name and enter it into the "Send to", section.
There could of been a message from someone that deleted their profile. This could give you a new message notification and the message isn't there any longer.

Assuming you are using the new version of Twitter, go to the top bar where it says "Messages". Once there, on the side it will show your message. Click it, and it opens up the conversation on the right side. If you are using the old Twitter, then go to the tab on the right side of your home page that says "Direct Messages" and it will display your messages down the page.

you click their username and it goes to their page. under it says send a message. click it and a message pops up.
