Every director has different methods of auditioning. Most will tell you to learn a certain piece of script, or prepare a song from a certain genre. You will then be given a time you will be auditioning, go into the audition room in front of the directors panel, and present what you have prepared. Some directors, however, will do a workshop audition. They consider this a lot less stressful for the auditionees and being a professional actress I have to agree. In most workshop auditions, they will do a certain amount of exercises and acting tasks and movement tasks and voice exercises with you. You will always be in a room with other auditionees and don't have to prepare anything. You just turn up and do what the board of directors tell you too. it depends on what your adjudicator wants you to do. you may have to cold read a few scenes with a partner, or you might have to memorize two monologues and perform them. if you are auditioning for a singing role, then you'll have to prepare a song, learn a dance you'll be taught at the audition and perform a scene.
first figure out the requirements. you will most likely need to memorize a monologue or two. (make sure that they are based off either the style of the show, or the mood of the character you wish to acquire.) after you memorize your monologue(s), if it's your first time auditioning, rehearse it in front of friends/family/etc. nothing is worse than stumbling/forgetting/mumbling over your words in an audition. (if you have braces on i suggest taking off your rubber bands. they tend to close the jaw)
if you are auditioning for musical theater, warm up your voice, eat/drink the appropriate foods and STRETCH!!!! (musical theater 90% of the time will include a dance portion of the audition) ALSO, if you're singing in your audition DO NOT DANCE AROUND THE STAGE UNLESS THEY ASK FOR IT! the singing portion is just to hear your voice, not to see if you can block a song well.
when preparing yourself for your monologue, if you find that you can't relate to it at all, just switch it. once you have found one that works for you, mold it. take the words and make it yours.
