Since they had to destroy a Horcrux they decided to get a basilisk fang from that place. Ron speaks in Parseltongue as heard from hearing Harry opening the Slytherin locket, and opens the chamber. Hermione stabs the Horcrux and there's a wave of water, then Ron and Hermione kiss.

He didn't - Hermione did. When Harry is searching for Ravenclaw's diadem Ron and Hermione go down to the Chamber of Secrets to get some Basilisk fangs (one of the few things that is able to kill a Horcrux). Hermione then stabs the cup and destroys it.

if your talking about the 7 th book, Hermione technically didnt get it, it was Ron but she went with him. They got it because Ron had watched harry speak parsel tongue the last time he opened the locket-horcrux. Ron then mimicked Harry to get him and Hermione into the chamber so they can get the basilisk fang to destroy the horcrux.(the basilisk was already dead when harry killed it in his second year.) Answer: from the dead Basilisk's mouth.
Have you even seen or read Harry Potter? In the Deathly Hallows part 1, when Harry opens the locket/horcrux ,so Ron can destroy it with Godrick Griffindor's sord, the horcrux fights back by showing Ron's worst nightmares wich are spiders, Harry and Hermione trash-talking about him, and Harry and Hermione kissing naked. But the scene doesn't show any inappropriate body parts, just Harry and Hermione's backs. (PS Hermione is played by Emma Watson.)
