Until the NCAA stripped Memphis of the 2008 season on August 20, 2009 Memphis held the record with a season of 38-1. Unless Memphis' appeal is successful all games in which Memphis fielded an inelligible player, Derrick Rose, will be cancelled resulting in the record being held by the following 4 teams:
Duke [2] (1986 (37-3)), UNLV 1987 (37-2), Duke [2] 1999 (37-2), Illinois [12] 2005 (37-2) and Kansas[1] (2008 (37-3))

The most wins in a single NCAA season is 37* - UNLV, Duke (twice), Illinois, and Kansas.

Kansas now claims the most wins in a Season, with a National Championship. Previously, each of the teams with 37 win seasons had failed to win the National Championship in their record season.
Really weird when you think about it.
*38 - Memphis, 2008 (38-2) Memphis' record 2008 season with 38 wins has been vacated by the NCAA for numerous rule violations.
*Note: Until it is OFFICIAL, Memphis will still hold this record. The University of Memphis is appealing the decision, so until all that is settled, the 38 wins is still it. IMO, if Duke didn't have to vacate it's wins and Final 4 in 1999 with Corey Maggette ADMITTING to taking money, then why does Memphis? Because they are in a non-BC$ league, that's why.
*Note: It is OFFICIAL. The wins have been vacated and Memphis is currently trying to get them back. **Derrick Rose was not an eligible player and there for should not have played for Memphis in the 2007-08 season. Any game he played in should have and was ruled a loss for Memphis.

University of Memphis. Memphis went 38-2 in the 2007-2008 season. They lost to Tennessee in the regular season, and Kansas in that epic National Championship game in which Memphis controlled the game, but blew a lead in the last 2 minutes. Not any more. Memphis was ruled in violation of NCAA rules that year, and has been forced to vacate their wins that season. So, effectively, they now went 0-40 that year.
The question is a little vague, but: UCLA has the most championships Kentucky has the most wins North Carolina is a close second in wins, and occasionally overtakes Kentucky.
Men's: Kentucky, 2011-12, 38-2
Women's: Baylor, 2011-12, 40-0

The Memphis Tigers hold the record for the most wins in a single season. In 2008, the Tigers went 38-2 for the season. I don't believe any other Division 1 mens basketball team has done that.
