There are a couple of options. If possible, you could drill the screw hole out to the next bigger size and tap it out to the next bigger size screw. Before you do this make sure you are not going to drill into a water jacket, oil port, or something else equally important. The next option is to have new threads heli-arced into the old hole. A quality machine shop will be able to do this for you.

Good luck removing a stripped screw from plastic. There are ways, but I'm not going to discuss because you'll probably burn your house down.
If the threads on the drain plug are stripped and the pan threads are good, you can just replace the drain plug. More often though, the threads in the oil pan that the drain plug screws into are what get stripped. If it's the oil pan, there are still a number of pretty simple options: There are oversized plugs you can buy, rethreading kits with inserts or self-tapping plugs. And there are rubber expanding plugs that are almost foolproof. It is your choice as to how you want to fix it. Go to your local auto parts store and look at your options. Whatever you do, do not use a silicone sealer to fix it.

Follow hose from radiator to engine. The hose at top of radiator near coolant reservoir tank. Where it connects to engine is where the thermostat is. There are two screws. Stripped mine. You know its the thermestat because the hose going there stays hard after running engine for a while, and you would be able to squeeze it if the thermostat opened up.
