The Quba Mosque. It is located outside Medina, Saudi Arabia. Iis the first mosque ever built and the oldest mosque of Saudi Arabia. Its first stones were positioned by the prophet Muhammad on his emigration from the city of Mecca to Medina and the mosque was completed by his companions. Muhammad spent more than 20 nights in this mosque (after migrating) praying qasr (a short prayer) while waiting for Ali whose house was behind this mosque.

There are two different answers depending on how this is to be read:

The first place that was consecrated as a Mosque was the Masjid al-Nabawi (The Prophet's Mosque) which was built in Medina, Saudi Arabia in 622 C.E.

The oldest building/structure which currently serves as a Mosque is the Kaaba surrounded by the Grand Mosque (AlMasjid Alharam) located in Makkah (or Mecca) in Saudi Arabia, because it was built by Abraham.

The house of worship for Muslims is the local Masjid or Mosque. The most important of the place of worship are the Kabbah in Makkah, the Prophet's Masjid in Madina in Saudi Arabia, and the famous Al Aqsa Masjid in Jerusalem.
