If you are talking about his 1936 production for the Federal Theatre Project, the big giveaway was the palm trees on the backdrop and the nineteenth-century costumes, as well as the inclusion of other voodoo elements which gave that production the nickname "The Voodoo Macbeth" (only a few minutes of this production were filmed, but you can find it online). If you are talking about Welles's 1948 film, everything, and especially the Scottish accents assumed by Welles and Jeannette Nolan, tells us that the setting of the play has not been changed at all, but was intended to be Scotland.
