There were no allegations that Lollia Paulina was a mistress of Claudius. We have very little information about her because Roman writers only mentioned her a few times and only briefly. There is no mention of an affair with Claudius whatsoever.

Lollia Paulina is mentioned only once in Suetonius' Life of Caligula (The Lives of Twelve Caesars, Life of Caligula, 25) where he mentions her forced marriage to Caligula and once in his Life of Claudius (The Live of the Twelve Caesars, Life of Claudius, 26) where he just mentions that she was a candidate for marriage with Claudius. Cassius Dio mentioned her only twice and only said that she was forced to marry Caligula (Roman Histories, LIX, 11) and that she was "slayed" by Agrippina (Roman Histories, LXI, 32). Tacitus gives more detail, but he only mentioned Lollia Paulina three times () and only talked about how her candidacy for marriage with Claudius was turned down, that charges against her were levied, that Claudius summoned the senate to punish her, that she was forced to commit suicide (Annals XII, 1-2, 22) and that Nero arranged for her ashes to be returned and the erection of a tomb for her (Annals, XIV, 12)

In 38 Lollia Paulina was forced to divorce Publius Memmius Regulus, and to marry Caligula. In 39 or 40 Caligula divorced her and married Milonia Caesonia. Cassius Dio said that that Caligula began an affair with her prior to their marriage and that she gave birth to their daughter one month after their wedding (Roman History, LIX, 23, 28). Suetonius said that Caligula announced that he had married her on the day she gave birth. (The Lives of Twelve Caesars, Life of Caligula, 25). Caligula was assassinated in 41.

When Claudius' third wife, Messalina, died in 48, there was a search for candidates for a new wife, with fierce competition between eligible women. Lollia Paulina and Agrippina the Younger emerged as the top candidates. The latter won on the basis that her parentage would keep imperial power within the Julio-Claudian family. Agrippina immediately plotted for the demise of Lollia. In 49 she had Lollia accused of having engaged in forbidden consultations with occultists. Claudius, without hearing the accused, summoned the senate to confiscate her property and exile her. Tacitus said that a tribune was sent to force Lollia to commit suicide and suggested that this had been ordered by Agrippina (Annals XII, 22).

When Nero ordered that Lollia Paulina's ashes be returned, presumably it was known where she had been buried. Tacitus does not give any details about this.
