Stapleton killed Sir Charles Baskerville, by setting his hound on him. The dog was made up to look like a hellhound and it is heavily implied- if not blatently stated? that the dog was starved and possibly beaten. It didn't kill him, though- he saw it and he died of fright, which I think is what Stapleton intended.
