Anything that could possibly come up in conversation. However, that's a pain to do, and it's easier to just expose yourself to massive quantities of English--movies, newspapers, books, music...--and then learn to figure out words based on context.
Given that English is one of wordiest (yes, that is a word) languages on Earth, you'd be memorizing a lot of words to become fluent.

It is calculated that the average English speaker uses about 2000 - 4000 words in ordinary conversation.

What country has more English speakers than the US?

This would depend on what one means by "Speak English." Certainly many Chinese and Indians study English and by pure population advantage should have more "speakers," but this "speaking" is often quite limited in scope and certainly could never be considered fluent. To use the definition of "English" and fluent loosely, there is no country with more fluent English speakers than the US. Here's a list from Wikipedia <http://en.wikipedia.org/wiki/List_of_countries_by_English-speaking_population>

What are the differences between formal and informal English?

There may be special meanings of "formal" and "informal" English with which I am not familiar. Formal English would be "standard English", and would be of more importance in written communication. Standard English would be the use of vocabulary and grammar that is as free as possible from idiomatic, slang or otherwise specialized use that would not be universally accepted or understood by fluent speakers world-wide. Informal English would be the English that local communities use, with all the idiomatic, slangy and quirky richness that that implies.
