Computers work with immediate storage called RAM (Random Access Memory) and longer term storage called ROM.
When a Computer runs an Application, the Computer needs to remember stuff to run the application, such as what the program looks like. This is Graphics Memory, and one example of RAM.
I encourage you to look at the Wikipedia page for RAM (included in related links), this may provide more insight.
RAM - is fast, temporary memory. A hard-drive is slower, but permanent. It's quicker for a computer to 'load' data into RAM, than to leave it on the hard-drive. Aditionally, all data in RAM is lost when the power is switched off. The data on a hard drive stays there until it's wiped by the user.
Computers have both immediate access store and backing store so the computer can save a short term memory and a long term memory.

Databases allow businesses to store information about their employees and about their customers. With a database employees can access this information using computers instead of going to a warehouse.
a hard drive stores the computers information, while the RAM (random access memory) can store temporary things like your history etc that get deleted when you turn off your computer
