Check the spark plug and the wire on the 8th spark cylinder. Corosion, short, or just bad.
You have not given enough information to even begin to answer this question. If the truck won't start, What makes you think that it has a misfire? A misfire is typically one or more cylinders not firing when the truck is running. If you indeed have a dead cylinder, that alone will not keep the engine from starting , unless you have catastrophic failure,(Something broken inside the given cylinder). However if you are not getting any fire then the truck will not start. Also if the truck has recently run & it ran rough, (As in mis-firing), then it may need a tune-up , (Or something more involved), & if that condition is bad enough; then YES, it could keep the engine from starting.
Do a proper diagnosis to figure out the cause of the misfire, then repair it.
If you have a 4 cylinder engine, the number one cylinder is the one closest to the front of the truck. If you have a 6 cylinder, it the on the driver side closest to the front of the truck
Number 1 is the cylinder closest to the timing belt. Mind you: Cylinder #1 is not the first cylinder to fire in the firing sequence.
If the check engine light is blinking , that means a cylinder misfire has been detected
looking at the front of truck number 1 cylinder is on right handside right behind powersteering pump
Looking at the motor compartment from the front of the truck, the number one cylinder is the first on the left (passenger side)of the motor.
The cylinder numbers are front to back passenger side is 1,2,3,4, drivers side is 5,6,7,8.
