Yes. Since the story is an "allegory" it could easily work in a modern setting. An allegory means that the story isn't really about what happens, but what it means. So the story could be turned into a sci-fi story, or a fantasy story, or a detective story, and it would still have the same essential meaning.
The majority of modern critics see the story as an allegory for death. Prince Prospero and his friends are rich people that can afford to hide out and party while the countryside suffers from a great sickness. A person at the party wears a cloak and mask symbolizing the sickness outside. He walks from room to room until he is noticed by Prince Prospero, who is angry that the someone would remind the partiers of the death outside. Prospero is struck dead by the sight of the person in the Masque, followed closely by the other guests. The guest is a personification of death and disease.
The story can be seen as a metaphor for many things. Disease (particularly cancer, ebola, West Nile Virus, etc.). It can also be seen as a warning that even the rich and powerful will eventually succumb to the same problems that plague the poor.

While it is true that in Western society the color black is stronglyassociated with mouring and with death in general, the color red is as stronglyassociated with blood and bloody death (amongst other things, of course). The terrible illness (the Red Death) spoken of in the story is marked by the appearance of scarlet stains upon the face and body of the victim, and at the end of the story it is said that the revellers" dropped one by one in the blood-bedewed halls of their revel".So, in my opinion, the story is actually stronger because it is calledThe Masque of the Red Death.
