The character's voice was first performed by Margie Hines, and was later provided by several different voice actresses including Kate Wright, Ann Rothschild (a.k.a. Little Ann Little), Bonnie Poe, and most notably, Mae Questel who began in 1931 and continued with the role until 1938.

Betty Boop's voice and catch phrase (Boop-Boop-a-Doop) was based upon the performances of Helen Kane, a Broadway performer who was popular in the late 1920s and early 1930s. Helen Kane appeared in several early talkies (see clips on YouTube) and recorded twenty-two (78 RPM) recordings on RCA Victor between 1928-1930. These singles have recently been remastered and made available on CD as "The Original "Boop-Boop-a-Doop" Girl, Classic Years (1928-1930)".

The title of the digital CD notwithstanding, there are only two connections between Helen Kane and Betty Boop: 1. Betty Boop began as a caricature of Helen Kane, and 2. In 1932 Helen Kane sued Fleischer Studio (creator and distributor of Betty Boop Cartoons) for what today would be called infringement of intellectual property rights. Helen Kane lost that lawsuit.

Initially, several actresses were cast as the voice of Betty Boop when the character was launched in 1983. The animator of the character soon chose to use one actress, Mae Questel, exclusively. Mae continued to provide the voice for Betty Boop until 1939.
