No, Bella's father does not know she is a vampire. All he knows is that Bella has changed somehow. That is what Jacob told him. Charlie says he wants to know the least amount of information possible and only what's necessary.

In the Twilight series, Bella's dad, Charlie, eventually finds out that Bella is a vampire. It happens in the Breaking Dawn book when Bella reveals her transformation to him. Initially shocked, he eventually comes to terms with it and accepts her new life.
