I say, from personal experience that skunks of both genders can spray. I stumbled upon a pair of mating skunks (male and female) and upon observing their rituals (sex practices) they became agitated at my close presence. My video camera was probably bothering them - they discontinued skunk coitus and simultaneously sprayed me and my video camera. So, YES, skunks of both genders can spray and I would assume that skunks of mixed gender can also spray.

It is very difficult to tell the difference between a male and female skunk in the wild. Generally speaking, a male skunk can weigh as much as 3-4 pounds more than a female. The male skunk can also be longer by up to 2-3 inches. Female skunks will usually have a much longer and ornate tail than the males. The behavior is perhaps the best way to determine sex. A male skunk will be protective and assertive year round, whereas a female will avoid confrontation, except when protecting kits. During mating season, February - May, the male skunk will give off a scent to attract females. Female skunks will also make dens with other females during the winter for warmth.

One way to find out the difference is the tracks.. I know it isn't really the most useful way to find out but still.. The male's tracks are bigger, suggesting that the male is actually bigger than the female. Plus their paws are actually shaped differently.
