As Shylock so eloquently says, Antonio has abused him in public because of his employment (one of the few which Jews were allowed), insulted him and spit on him.

That's pretty rude, but it would appear that Antonio is not alone. Salerio and Solanio are both rough on Shylock, as is Gratiano. However, because Antonio is the one Shylock has a chance to get back at, Shylock intends that he should pay for all the abuse that Shylock has taken from all the Christians.

Antonio despises Shylock for being Jewish and Shylock hates Antonio for despising him. However, Antonio needs money, and Shylock is a moneylender. Antonio therefore goes to Shylock to borrow money, and Shylock is at first reluctant, but afterwards agrees in the hope that Antonio will default so he can take his pound of flesh.

Shylock offered to lend Antonio 3000 ducats without interest for three months, the only condition being that if Antonio failed to repay on the appointed day, Shylock could take a pound of flesh from any part of Antonio's body he chose.

Shylock sets a trap for Antonio by including a default clause in his contract which, if Antonio does not repay, could lead to his death. He further shows it by refusing to abandon the effect of that clause. But Shylock is not motivated only by hatred for Antonio. He is also angry at Lorenzo, who has stolen his daughter and his money and his wife's wedding ring, and is a spendthrift wastrel. But he cannot do anything to Lorenzo or any of the other Christians who have treated him badly. The only one he can get his hands on (he thinks) is Antonio, and he will make him pay for all the harm everyone has done to Shylock.
