It's very easy. You take the Uptown C or E trains(of the A-C-E, the blue line) from 34th Street (Penn Station) to 50th Street. It's only two stops: 34th Street ---> 42nd Street ---> 50th Street). The 50th Street Station on the C and E trains is at 50th Street and 8th Avenue.
Don't take the A, as it is an express train and will not stop at 50th Street.

Then walk 3 blocks south to 50th Street, turn left, and walk 1/2 a block east to the Kimberly Hotel, at 145 East 50th Street between Lexington and 3rd Avenues.
