It depends on how you mean "do" in English.

If you mean it purely as a placeholder for a different verb, such as the below conversation, you should say, "Me alegro que si"

I work very hard at my job.

I am glad you do. -- i.e. I am glad you work hard.

If you mean it in the sense of performing, i.e. the usual use of "do", then a translation would be: Me alegro que lo hagas.

It depends how this intended.

If you mean it in the sense that you are happy that the person does something, such as in the example below, use: Me alegra que lo haga / hagan/ hagas/ hagáis.

Person 1: As promised, I delivered your friends home safely. -- Como lo prometí, llevé seguros a todos tus amigos a su casa / a sus casas.

Person 2: I am glad you do this [service]. -- Me alegra que lo hicieras / hicieses (requires preterite subjunctive).

If you mean it to affirm a previous statement and the "do" refers to a verb other than "making" or doing, such as in the below, use: Me alegro que sí.

Person 1: I really like the food at this restaurant. -- Me gusta la comida de este restaurante, de verdad.

Person 2: I am glad you do. -- Me alegro que sí.
