Estrogen is produced in the ovaries, primarily the theca (wall) of developing follicles in the ovary, though also to a lesser extent the corpus luteum (remaining outer 'shell' which previously contained an egg) and, during certain stages of pregnancy, the placenta.

Some estrogens are produced in smaller quantities by liver, adrenal glands and the breasts.These secondary sources of estrogens are especially important in postmenopausal women. Fat cells produce estrogen as well.

Estrogen is a generic term for a group of steroidogenic hormones that includes estradiol, estrone, and estriol. Estradiol is produced by the ovary and is the primary and most abundant estrogen. Estrone is produced by both the ovary and adipose tissue. Estriol is produced by the placenta during pregnancy. All three are derived from metabolic products of cholesterol. Estradiol and estrone are derived from androstenedione, which is then either de-methylated and converted to estrone or converted to testosterone, which is further modified by aromatase into estradiol. Estriol is synthesized by the placenta from 16-OH DHEAS, another metabolic product of cholesterol.
