Nicotine has a half-life of about 2 hours, meaning, half of your nicotine dosis is out of your system within two hours, to get all substances out of your system, it takes about three days. (varies from person to person of course)

The metabolite of nicotine, called cotinine, is what is tested in the blood. The half-life of cotinine is 20-24 hrs, and up to five half-lives can be detected. This means that you would have to stop smoking cigarettes (and the use of any other nicotine containing substances) for about a week, for cotinine levels to be zero.
