Depends on whether you are talking about his immediate family or his extended one and when. His immediate family at the time of his death consisted of himself and his wife, two daughters and two sons-in-law and a granddaughter. His extended family consisted of his immediate family and his sister Joan and three of her children: William, Thomas and Michael.

But if you ask how many family members did Shakespeare have in 1595, you would have to count in his immediate family, his wife, son and two daughters, and in his extended family, his immediate family, his father, his mother, his brothers Gilbert, Richard and Edmund, and his sister Joan.
