Old fortinbras, Yorick, and King Hamlet are all dead at the start of the play. During the play itself, Polonius is killed, followed by Rosencrantz and Guildenstern (offstage), Ophelia, Gertrude, Claudius, Laertes, and finally Hamlet.

The first to die within the actual plot-line of the play is Polonius, father of Ophelia and adviser to the King of Denmark. However central to the themes and plot of the play was the murder by poisoning of Old Hamlet, the current Prince Hamlet's father and previous King of Denmark, and brother to Claudius.
In just that order. Polonius dies in Act 3 Scene 4, Ophelia dies in Act 4 Scene 7, and Claudius and Hamlet both die in Act 5 Scene 2. Hamlet kills Claudius then dies of the poisoned wound Laertes gave him.
