Not really! It's 16 hours on a direct route (by bus it could be a little longer). The distance between the two cities is only around 700 miles (1126 km), but the terrain is really windy and mountainous once you get past Ica en route.

Alternatively, there is the slightly safer route via Arequippa, but this takes an extra two hours (as it is further) and is not as scenic (though Arequippa is very pretty and worth visiting itself).

Also, you can fly between them there in an hour and 15, but the altitude sickness effect will be more acute, so be careful (Lima is at sea-level, whilst Cusco is at around 3300 metres!
